The StarLighter dsDNA HS Assay Kit is a simple, sensitive, and accurate fluorescent quantitative detection kit for double-stranded DNA (dsDNA). Suitable for use with Qubit fluorometers or fluorescent microplate readers, this kit exhibits excellent linearity for dsDNA in the range of 0.1-100 ng. It exhibits strong anti-interference capabilities, allowing selective detection of dsDNA even in the presence of ssDNA, RNA, and monomeric nucleotides, and is tolerant to high concentrations of contaminants such as proteins, salts, and detergents. Compared to traditional UV detection (A260) methods, it allows for more accurate DNA quantification. Its high specificity and sensitivity make it particularly suitable for quantitative detection of libraries during NGS library construction.

Life Qubit Standards 1 and 2 were used as test samples, and QB buffer was used as the diluent. A gradient series of 10, 5, 2.5, 1.25, 0.625, 0.3125, 0.156, 0.0781, and 0 ng/µL was prepared. The test system consisted of 10 µL sample and 190 µL working buffer. Nine dsDNA samples of varying concentrations were assayed using StarLighter Qubit reagent and Thermo Qubit reagent, respectively. The results are shown in Figure 1. The StarLighter dsDNA HS Assay Kit FS-T1001, FS-T1002, and the Qubit® dsDNA HS Assay Kit (Life) demonstrated comparable performance, exhibiting excellent linearity from 0 to 100 ng of dsDNA and consistent sample reproducibility. There was no significant difference in performance between the two assays.
