The dyes used in RT-PCR and SYBR Green qPCR mix do not affect reverse transcription and fluorescence quantification results. 40 ng and 1 ng of mouse total RNA were used as templates for reverse transcription using FS-P1005 and FS-P1006, respectively. The resulting cDNA was then quantified by qPCR using FS-Q1005 and FS-Q1008 for ATP and FMR1 genes, respectively. The dye-labeled system (FS-P1006 + FS-Q1008) performed similarly to the system without dye labeling (FS-P1005 + FS-Q1005), with no significant differences in amplification linearity, fluorescence intensity, or Ct values. |
