The proportion of ribosomal RNA (rRNA) in the total RNA of plants, animals and microorganisms can be as high as 90% or more, which greatly limits the detection efficiency and sensitivity of transcriptomes (such as mRNAs) in RNA-seq. In RNA-seq experiments, efficient and specific removal of rRNA can save sample sequencing volume and effectively improve the detection sensitivity of target RNA.

The SP-ribo-PooLs ribosomal RNA removal series products developed by Qihengxing Biotechnology, an expert in the development of ribosomal RNA (rRNA) removal reagents, have been carefully calculated and optimized to design a highly complex biotin-modified DNA probe pool that can efficiently and specifically bind to rRNA to obtain unbiased RNA-seq results.

1. Product Introduction

Single Species ribo-Pools are used to study known and lesser-known species (such as humans, house mice, brown rats or chinchillas, etc.). SP-ribo-Pool for Single Species is carefully calculated and complexly designed based on the rRNA of the species. The probes in the probe pool can evenly cover the 5S, 5.8S, 18S, 28S rRNAs in the cytoplasm and the 12S, 16S mitochondrial rRNAs.

Among them, the key indicators of probe design include:

• Meets criteria for mismatch with whole genome

• GC% range required at optimal hybridization temperature

Agilent Bioanalyzer analysis results of rRNA-depleted samples

Total RNA (RIN ≥ 7.5) extracted from HeLa cells or mouse embryonic fibroblasts was used at an input of 1 μg for rRNA depletion using the Human/Rat/Mouse SP-ribo-Pool and Ribo-Zero Gold (Human, Mouse, Rabbit) kits (Illumina). The Human/Rat/Mouse SP-ribo-Pool kit performed better or equivalent to the Ribo-Zero Gold kit in removing 18S/28S ribosomal RNA. The removal of RNA from human samples was equivalent to that of RNA from mouse samples.


RNA sequencing showed that SP-ribo-Pool was more efficient in removing rRNAs than the Ribo-Zero Gold kit.

For human and mouse samples, the SP-ribo-Pool kit showed approximately 4% higher rRNA removal efficiency than Ribo-Zero Gold.
