DNA was extracted from samples from different locations and soil types using the Qihengxing Soil DNA Extraction Reagent and various imported and domestic soil DNA extraction kits, with triplicates performed for each sample. For most samples, the Qihengxing StarPure Soil DNA Extraction Kit extracted higher amounts of DNA than kits from other suppliers. |

Different numbers of rice blast fungus spores were added to sterile soil to achieve final concentrations of 5,000, 10,000, 25,000, 50,000, 100,000, 250,000, and 500,000 spores per 500 mg of soil, respectively. DNA was extracted from these soils using Qihengxing, imported, and domestic brand soil extraction kits. 50 μl of elution buffer was used for elution, and 4 μl of the extracted DNA was used for qPCR detection using Qihengxing StarLighter SYBR Green qPCR Mix (FS-Q1002). The Ct values are shown in Table 1. The Qihengxing soil extraction kit demonstrated excellent extraction efficiency and better input-output correlation (higher R 2 ). |
