Accurate library quantification is very important for the results of second-generation sequencing. Underestimating the library concentration will lead to excessive cluster generation and poor data quality; overestimating the library concentration will lead to low data output and low genome coverage. Therefore, underestimating or overestimating the library concentration will affect the sequencing results. qPCR is the gold standard for library quantification and can accurately detect the number of molecules in the library. This is because only library fragments with adapters at both ends will be amplified and accurately quantified. Fragments without adapters or with adapters at only one end cannot be amplified, and these fragments will not be quantified; while other quantitative methods, such as Qubit, Nanodrop and other methods, will quantify all fragments, resulting in deviations in the quantitative results.
The kit includes the qPCR mix, primer pairs, and six concentrations of standards required for library quantification.

    Tab.2: Both Supplier K and the Qihengxing Library Quantification Kit contain six standards, offering a wide quantification range. Six standards from different batches of Supplier K and the Qihengxing Library Quantification Kit were used as samples and amplified using the same qPCR reagent. The amplification results were then subtracted. Comparison revealed minimal differences between the standards in the two kits, demonstrating comparable performance. The results are shown in the table above.
    2. High amplification efficiency and accurate quantitative results
    Fig. 2. Competitor standards S4 and S6 were used as samples and amplified using the Qihengxing and competitor kit mixes, respectively. The amplification results were compared with the standard standards in the kit. The quantification results from the Qihengxing and competitor kits were both close to the labeled concentrations. The Qihengxing quantification kit provided accurate and reliable library quantification results.
    Tab.3: Standards S4 and S6 from Supplier K were used as samples for amplification and quantification using the Qihengxing and Mix kits from Supplier K, respectively. The quantitative results were compared with the labeled concentrations of the standards in Supplier K's kit. The quantitative results from both the Qihengxing and Supplier K kits were close to the labeled concentrations. The Qihengxing quantification kit provided accurate and reliable library quantification results.
