Accurate library quantification is very important for the results of next-generation sequencing. Underestimating the library concentration will result in excessive generation and poor data quality; overestimating the library concentration will result in low data output and low genome coverage. Therefore, underestimating or overestimating the library concentration will affect the sequencing effect. qPCR is the standard for DNA library quantification and the only method that can accurately detect the number of molecules.

    The concentration of the library to be tested is calculated by absolute quantification method and used for library determination before NGS sequencing.

    Tab.2 Both Supplier K and Qihengxing Library Quantification Kit contain 6 standards, with a wide quantitative range. Six standards from different batches of Supplier K and Qihengxing Library Quantification Kit were taken as samples, amplified using the same qPCR Mù, and the amplification results were subtracted. The difference between the standards of the two kits was minimal, and the effects were equivalent. The specific results are shown in the table above.
    2. High amplification efficiency and accurate quantitative results
    Fig.2 Take the standard products S4 and S6 in the competitor's kit as samples, and amplify them with the Mix in the Qihengxing and competitor's kits respectively. The amplified results are compared with the quantitative results of the Qihengxing kit for the standard products in the kit and the quantitative results of the competitor's kit, both of which are close to the labeled concentration. The library quantification results of the Qihengxing quantitative kit are accurate and reliable.
    Tab.3 Take the standard products S4 and S6 from supplier K as samples, and use the Mix in the Qihengxing and supplier K kits for amplification quantification, respectively. The quantitative results are compared with the standard products labeled in the supplier K kit. The quantitative results of the Qihengxing kit and the supplier K kit are close to the labeled concentration. The library quantification results of the Qihengxing quantitative kit are accurate and reliable.
