StarLighter Color Script RT super mix(+gDNAremoval) The reverse transcription mix developed using the latest generation of reverse transcriptase (fourth-generation genetically engineered enzyme) contains the reverse transcriptase, random primers, Oligo dT and other reaction components required for reverse transcription. The reverse transcription reaction can be carried out by directly adding the RNA template to the system. At the same time, the mix contains a visible dye to facilitate the visualization of sample addition. In addition, the kit is also equipped with gDNA removal and its buffer for efficient gDNA removal, which can effectively remove the impact of gDNA contamination on qPCR quantitative results during reverse transcription. The latest generation of reverse transcriptase is resistant to high temperatures and can exhibit good reverse transcription activity at temperatures of 42"C-65"℃, and can effectively reverse transcribe RNA with complex structures.

Reverse transcription was performed using 40 ng and 1 ng of mouse total RNA as template using StarLighter Color Script RT (FS-P1006) and StarLighter Script RT (FS-P1005), respectively. The resulting cDNA was then quantified by qPCR for ATP and FMR1 using StarLighter HP SYBR Green qPCR Mix (FS-Q1005). FS-P1006 and FS-P1005 showed comparable performance, with no significant differences in amplification linearity, fluorescence intensity, or Ct values.

StarLighter Color HP SYBR Green qPCR Mix (FS-Q1008) contains a blue dye. cDNA samples reverse-transcribed with StarLighter Color Script RT Super Mix are yellow. When the StarLighter Color HP SYBR Green qPCR Mix is added to the yellow cDNA template, it turns green, enabling tracking of sample addition.

  • The dyes in RT and SYBR mix do not affect reverse transcription and quantification results.


Reverse transcription was performed using 40 ng and 1 ng of mouse total RNA as template using StarLighter Script RT (FS-P1005) and StarLighter Color Script RT (FS-P1006), respectively. ATP and FMR1 gene expression was quantified by qPCR using StarLighter HP SYBR Green qPCR Mix (FS-Q1005) and StarLighter Color HP SYBR Green qPCR Mix, respectively. The dye-labeled RT plus the dye-labeled qPCR mix (FS-P1006 + FS-Q1008) performed identically to the dye-free RT plus the dye-free qPCR mix (FS-P1005 + FS-Q1005). Amplification linearity, fluorescence intensity, and Ct values were similar.
