StarLighter Color HP SYBR Green qPCR Mix This qPCR reagent contains a genetically engineered enzyme that tolerates high concentrations of SYBR Green I dye. Adding sufficient SYBR Green I dye to the reaction system can reduce the number of cycles required for the fluorescence signal to reach threshold (Ct value), thereby improving signal-to-noise ratio, linearity, and sensitivity. The optimized reaction system enhances amplification specificity, effectively reducing nonspecific amplification and providing more reliable data.

StarLighter Color HP SYBR Green qPCR Mix contains a blue dye, and 5× Sample Diluent contains a yellow dye. When a sample diluted with Sample Diluent (yellow) is added to StarLighter Color HP SYBR Green qPCR Mix (blue), a color change from blue to green occurs, allowing accurate identification of template addition based on the system color.

  • Dyes do not affect the performance of the reagents



Reverse transcription was performed using 40 ng and 1 ng of mouse total RNA as template using StarLighter Color Script RT (FS-P1006) and StarLighter Script RT (FS-P1005), respectively. The resulting cDNA was then quantified by qPCR for ATP and FMR1 using StarLighter HP SYBR Green qPCR Mix (FS-Q1005). FS-P1008 and FS-P1005 showed comparable performance, with no significant differences in amplification linearity, fluorescence intensity, or Ct values.

  • Combined with StarLighter Template Tracer High-Performance Dye-Based qPCR Master Mix (FS-Q1008), it enables sample addition and tracing.



StarLighter Color HP SYBR Green qPCR Mix (FS-Q1008) contains a blue dye. cDNA samples reverse-transcribed with StarLighter Color Script RT Super Mix are yellow. When the StarLighter Color HP SYBR Green qPCR Mix is added to the yellow cDNA template, it turns green, enabling tracking of sample addition.
  • The dyes in RT and SYBR mix do not affect reverse transcription and quantification results.

ATP and FMR1 gene expression was quantified by qPCR using StarLighter HP SYBR Green qPCR Mix (FS-Q1005) and StarLighter Color HP SYBR Green qPCR Mix, respectively. The dye-labeled RT plus the dye-labeled qPCR mix (FS-P1006 + FS-Q1008) performed identically to the dye-free RT plus the dye-free qPCR mix (FS-P1005 + FS-Q1005). Amplification linearity, fluorescence intensity, and Ct values were similar.
